Following the start in Glenelg, the riders head south down the Southern Expressway. They move through Old Noarlunga to head for the only classified uphill of the day. Sellicks Hill is crested at kilometre 43.9 after a 5.4 kilometres climb at 2.3%. The route continues to Myponga before veering east towards the south-coast of the Fleurieu Peninsula. For 20 kilometres the riders race from one seaside town to the next. After Victor Harbor, Port Elliott, Middleton and Goolwa the Tour Down Under is once again an area of gently sloping hills for the finish in Strathalbyn.
The first three riders on the line take time bonuses of 10, 6 and 4 seconds, while the intermediate sprints (at kilometre 47.2 and at kilometre 73.9) come with 3, 2 and 1 seconds.
